醫(yī)學論文范文:川芎嗪對谷氨酸損傷海馬神經(jīng)元的保護作用
【摘要】 目的 觀察川芎嗪對谷氨酸損傷海馬神經(jīng)元的保護作用。方法 原代培養(yǎng)大鼠海馬神經(jīng)元,培養(yǎng)基中加入1mmol/L的鹽酸川芎嗪溶液共同孵育12h后,加入1mmol/L谷氨酸損傷海馬神經(jīng)元20min。采用MTT法檢測細胞活性;測定培養(yǎng)液中乳酸脫氫酶(LDH )活力的變化;細胞免疫組化檢測各組細胞表達AchE。 結果 川芎嗪可提高谷氨酸損傷后海馬神經(jīng)元的存活率,抑制因谷氨酸損傷引起的細胞內(nèi)LDH的釋放,可促進谷氨酸損傷細胞中AChE的表達。 結論 川芎嗪對谷氨酸誘導海馬神經(jīng)元損傷有保護作用。

Soochow University, Suzhou 215123, China)ABSTRACT: Objective To observe the protective effects of ligustrazine on glutamateinduced injury in cultured hippocampal neurons. Methods Primarily cultured hippocampal neurons from fetal rats were incubated with ligustrazine (1mmol/L) for 12 hours, then glutamate (1mmol/L) was added for 20 minutes to induce injury. Cell viability was detected by MTT assay, and the change in LDH activity was determined by biochemical method. Finally, the expression of AchE in the cultured neurons was assayed by immuocytochemistry. Results Ligustrazine improved the survival rate of hippocampal neurons injured by glutamate, inhibited the release of LDH from the kytoplasm injured by glutamate. The expression of AchE in the injured neurons was promoted by pretreatment of ligustrazine. Conclusion Ligustrazine can significantly exert protective effects on the hippocampal neuron injury induced by glutamate.

海馬是邊緣系統(tǒng)的重要組成部分,參與學習、記憶、情緒反應及植物神經(jīng)功能等。原代培養(yǎng)的海馬神經(jīng)元對許多致病因素和化學損害較為敏感,是研究神經(jīng)元損傷及抗損傷的理想模型。在腦缺血模型中,海馬神經(jīng)元的損傷較為嚴重,其中興奮性氨基酸含量增高是海馬神經(jīng)元損傷的原因之一。谷氨酸(glutamate, Glu)是哺乳動物腦內(nèi)主要的興奮性氨基酸,參與中樞神經(jīng)系統(tǒng)的信息傳遞。但Glu過度釋放可產(chǎn)生神經(jīng)興奮性毒性,引起神經(jīng)細胞損傷或死亡。川芎嗪(ligustrazine, Lig)是從中藥川芎總生物堿中分離得到的一種有效活性生物堿。Lig能通過血腦屏障進入中樞神經(jīng)系統(tǒng),在腦中快速達到峰值,并持久地存留在大腦內(nèi)。臨床上常用Lig治療腦缺血,其機制是通過擴張微血管改善微循環(huán),抑制血栓形成,阻斷自由基連鎖反應,降低細胞內(nèi)鈣離子超載,拮抗氨基酸毒性等途徑來實現(xiàn),具有多靶點、多層次、多環(huán)節(jié)的腦保護作用[1]。本研究采用體外海馬神經(jīng)元原代培養(yǎng),觀察Lig對Glu損傷海馬神經(jīng)元的保護作用,探討Lig對腦損傷神經(jīng)元的保護機制。
