本發(fā)明涉及生物基因領(lǐng)域,具體涉及日本血吸蟲抱雌溝蛋白的基因序列及在大腸桿菌中的表達(dá)。本發(fā)明根據(jù)已報(bào)道的曼氏血吸蟲編碼抱雌溝蛋白的基因SmGcp序列和編碼日本血吸蟲抱雌溝蛋白保守區(qū)的cDNA序列SjGcp1設(shè)計(jì)兩對(duì)引物,分別擴(kuò)增出日本血吸蟲抱雌溝蛋白編碼序列的5’和3’區(qū)段,再根據(jù)測(cè)序結(jié)果設(shè)計(jì)編碼全長(zhǎng)引物,從蟲體中抽提總RNA,用PCR克隆出該基因,克隆后經(jīng)全序列測(cè)定表明該成熟蛋白cDNA編碼1932個(gè)核苷酸,即643個(gè)氨基酸。該基因的成功克隆及基因序列測(cè)定為以后闡明血吸蟲的生殖發(fā)育機(jī)理,以及從阻滯雌蟲成熟方面防治血吸蟲病打下了良好的基礎(chǔ)。
