醫(yī)學論文范文:辛伐他汀對高糖誘導的腎小管上皮細胞轉(zhuǎn)分化的影響
【摘要】 目的:觀察辛伐他汀對高糖誘導的腎小管上皮細胞轉(zhuǎn)分化的作用。方法:大鼠腎小管上皮細胞隨機分成三組(n=8):對照組,高糖組和辛伐他汀組。對照組加培養(yǎng)液,高糖和辛伐他汀組在培養(yǎng)液中加葡萄糖,終濃度為30 mmol/L,辛伐他汀組加入終濃度為1 μmol/L辛伐他汀。在6,12,24,48,72 h時用免疫細胞化學和Western blot檢測α-SMA的表達。結(jié)果:對照組各時間段均無α-SMA表達。高糖組6 h時出現(xiàn)α-SMA陽性表達,48 h達高峰。與高糖組比較,辛伐他汀組各時間點α-SMA的表達均明顯減少(P<0.05)。結(jié)論:辛伐他汀可抑制高糖誘導的大鼠腎小管上皮細胞轉(zhuǎn)分化。

Effects of Simvastatin on the Transdifferentiation of Renal Tubular Epithelial Cells Induced with High Glucose UANG Xiao-xia,KE Chang-bin,ZHANG Qing-hong,ZHANG Jian-e^ (Department of Nephrology;Department of Anesthesia,Taihe Hospital,Yunyang Medical College,Shiyan,Hubei 442000,China)Abstract:Objective To investigate the effects of simvastatin in renal tubular epithelial cells transdifferentiation induced with high glucose.Methods Cultured renal tubular epithelial cells were randomly divided into three groups(n=8):control group(group C),high glucose group (group D) and simvastatin group(group S),and given culture solution,high glucose(terminal concentration:30 mmoL/L),high glucose(terminal concentration:30 mmoL/L) plus simvastatin(1 μmoL/L), respectively. The expression of α-SMA was detected with immuocytochemistry and Western blot at 6,12,24,48 and 72 hours,respectively.Results The expression of α-SMA was not detected at all the time points.The positive expression of α-SMA in group D was detected at 6 h and reached peak at 48 h.The expression of α-SMA in group S was significantly lower than that of group D at all the time points(P<0.05).Conclusion Simvastatin could inhibit the transdifferentiation of renal tubular epithelial cells induced with high glucose醫(yī).學全.在.線網(wǎng)站gydjdsj.org.cn.

糖尿病腎病腎間質(zhì)纖維化并進展成終末期腎衰竭是糖尿病危及患者生命的重要原因,其中肌成纖維細胞(Myof)大量增生并分泌平滑肌肌動蛋白(α-smooth muscle actin,α-SMA)是其中心環(huán)節(jié)[1-2]。辛伐他汀有抗組織纖維化[3-5],改善腎小球硬化、腎臟保護作用,能延緩腎臟病變的進展[6-11]。但是否能夠通過抑制腎小管上皮細胞轉(zhuǎn)分化而抑制腎纖維化報道甚少,本文觀察辛伐他汀對高糖誘導的腎小管上皮細胞轉(zhuǎn)分化的作用。
1 材料和方法
1.1 主要試劑
α-SMA單克隆抗體(武漢博士德生物工程有限公司),α-SMA免疫細胞化學用二抗、Western blot用HRP標記二抗(北京中杉金橋生物技術(shù)有限公司)。
1.2 大鼠腎小管上皮細胞的培養(yǎng)
大鼠腎小管上皮細胞株(NRK-52E)購于ATCC。用含5%胎牛血清的DMEM培養(yǎng)基,在5%CO2、37 ℃的條件下培養(yǎng)。
